Key findings
- TRIUMPH-1 (Phase 3; adults with obesity/overweight + ≥1 weight-related comorbidity, without diabetes): at 80 weeks, average weight loss 19.0% (4 mg), 25.9% (9 mg), 28.3% (12 mg) vs 2.2% placebo (treatment-regimen estimand; efficacy estimand up to ~30.3%). 45.3% of 12 mg patients lost ≥30% of body weight — a level long associated with bariatric surgery (~70.3 lb at 12 mg).
- TRIUMPH-4 (first Phase 3 readout, Dec 2025): up to 28.7% weight loss (~32.3 kg / 71.2 lb) at 68 weeks, 75.8% reduction in osteoarthritis pain, ~20% LDL reduction, 72% reversal of prediabetes to normoglycemia.

Why it matters for this base
- Existing KB evidence for Retatrutide was Phase 2 (Jastreboff 2023, ~24% at 48 wk) plus transcripts. This is the first Phase 3 confirmation, and the first to add osteoarthritis pain and cardiometabolic endpoints — the strongest GLP-class weight-loss data to date.
